Description
MOTS-c – Characteristics, Structure, and Applications in Laboratory Research
What is MOTS-c?
MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA Type-c) is a mitochondrial-derived peptide (MDP – Mitochondria-Derived Peptide), discovered in 2015. Unlike most known peptides and proteins, which are encoded by nuclear DNA, MOTS-c is formed based on information stored in mitochondrial DNA (mtDNA), within the gene encoding mitochondrial 12S rRNA. This unique feature has made the peptide a subject of intensive research in mitochondrial biology, molecular biology, and cellular metabolism.
Since its discovery, MOTS-c has been widely used in research on communication between mitochondria and the cell nucleus, as well as the mechanisms of cellular response to metabolic stress. This compound belongs to a relatively new group of mitochondrial peptides, which also includes Humanin and SHLP (Small Humanin-Like Peptides).
Chemical Structure
MOTS-c is a synthetic analog of a naturally occurring peptide composed of 16 amino acids. It corresponds to the sequence encoded by a small open reading frame (sORF) located within the mitochondrial 12S rRNA gene.
Unlike most research peptides whose genes are found in the nuclear genome, MOTS-c represents a class of molecules encoded by the mitochondrial genome. For this reason, it is often used as a research model in analyzing the dependencies between mitochondrial function, gene expression, and cellular signaling.
MOTS-c as a Subject of Scientific Research
MOTS-c is one of the best-understood mitochondrial-derived peptides. Scientific publications dedicated to this molecule primarily focus on its role in cellular signaling and mechanisms regulating cellular energy homeostasis.
The most frequently described research areas include:

Due to its unique origin, MOTS-c is often used as a reference compound in studies on mitochondrial DNA-encoded peptides and the analysis of their molecular properties.
Significance in Peptide Research
MOTS-c is one of the most important mitochondrial-derived peptides described in scientific literature. Its discovery changed the perception of mitochondria, indicating that they not only function in energy production but also participate in transmitting signals regulating cell function.
For this reason, MOTS-c remains the subject of numerous studies concerning mitochondrial biology, cellular signaling, gene expression regulation, and the design of new peptides inspired by natural molecules encoded by the mitochondrial genome.
